Topic: A new mechanism of genome instability

Abstract

Large insertions at DNA double-strand breaks (DSBs) threaten genome integrity and are common in cancer. Moreover, they also drive genome evolution by generating gene duplication. It is not clear how cells prevent this type of genome instability. We identified the first mutant in yeast or any organism with a phenotype of frequent insertions of ~0.1-1.5 kb long sequences into DSBs. This mutant is deficient in Dna2 nuclease. We propose a model that Dna2 prevents the formation of extrachromosomal DNA fragments and their insertion at DSBs.
